#986 Expression of the Truncated Somatostatin Receptor Variant Sstr5TMD4 in Medullary Thyroid Carcinoma
Introduction: Medullary thyroid carcinoma (MTC) is a rare tumor originating from thyroid parafollicular C cell. The only available treatment is surgical resection, which is not always feasible. Somatostatin and its receptors are expressed both in normal human thyroid C cells and in MTC, but their role has not been fully elucidated. The discovery of a truncated isoform of Sstr5, Sstr5TMD4, associated with increased invasiveness and aggressiveness in breast cancer, has opened new insight in the role of Sstr in cancers.

#1566 Splicing Dysregulation Impacts on Neuroendocrine Tumors: Evidence from Altered Spliceosoma Components and Somatostatin and Ghrelin Systems.
Introduction: Splicing is an emerging cancer hallmark influencing multiple tumor cell (dys)functions. We have shown that aberrantly spliced variants of somatostatin receptor subtype 5 (sst5TMD4) and of the pleiotropic neuropeptide ghrelin (in1ghrelin) are linked to poor prognosis in gastroenteropancreatic neuroendocrine tumors (GEPNETs) and may enhance their aggressiveness features.

#2141 Potential Role of SST5TMD4-Derived Peptides in the Malignancy of Neuroendocrine Tumors and Other Endocrine-Related Cancer Cells
Introduction: Extracellular fragments derived from membrane receptors can play relevant functional roles in certain tumoral pathologies and could provide novel diagnostic or therapeutic tools. We have identified a novel truncated somatostatin receptor type 5 variant, sst5TMD4, which is overexpressed and functionally active in different endocrine-related cancers, including neuroendocrine tumors (NETs). Interestingly, sst5TMD4 has 4 transmembrane domains and its C-terminal tail is consequently exposed towards the extracellular matrix, and could, therefore, be the substrate for proteolytic enzymes.
