#2944 Soluble ST2 (sST2) Levels in the Management of Carcinoid Heart Disease in Patients with Neuroendocrine Cancer
Introduction: The identification of a sensitive biomarker to detect the presence and severity of carcinoid heart disease could allow more selective use of clinical and echocardiographic screening. ST2 is a protein belonging to the family of interleukin 1 receptors, present both in the isoform of transmembrane (ST2L) and in the soluble one (sST2). sST2 blocks the anti-inflammatory, anti-hypertrophic, and anti-fibrotic effects of cytokine IL-33 on the myocardium and recent studies have shown that sST2 levels increase in the blood of patients diagnosed with heart failure.

#2853 Succinate Dehydrogenase B (SDHB) Immunohistochemical Expression in Pancreatic Neuroendocrine Tumours and Correlation with Somatostatin Receptor Immunohistochemistry
Introduction: The established category of succinate dehydrogenase (SDH) deficient neoplasms includes several endocrine tumours with recent evidence that pancreatic neuroendocrine tumours (PNETs) could be included in this tumour group. The SDH complex is composed of four subunits (A-D) but loss of immunohistochemical (IHC) expression for SDHB indicates bi-allelic inactivation of any SDH component and is used as a marker for syndromic disease.

#19 Antitumor activity of Pasireotide (SOM230) alone and in combination with Everolimus (RAD001) in DU-145 human prostate cancer model
Introduction: Pasireotide (SOM230) is a novel multi-receptor ligand somatostatin analogue with high affinity for somatostatin receptor subtypes sst1,2,3 and sst5. Like octreotide, which binds primarily to sst2, it inhibits hypersecretion of hormones from patients with functional pituitary tumors and gastroenteropancreatic neuroendocrine (GEP/NET) tumors. In addition, tumor shrinkage has been observed with both compounds in patients with acromegaly, Cushing’s disease and GEP/NETs, but its tumor-reducing mechanism of action has so far not been revealed. In patients with breast and liver cancer, octreotide had little or no antitumor activity.

#62 Genome-wide DNA methylation profiling of pancreatic neuroendocrine tumors identifies distinct methylation profiles and differentially methylated gene promoter regions associated with low, medium and high grade tumors
Introduction: Integration of genetics and epigenetics has emerged as a powerful approach to studying cellular differentiation (Mikkelsen et al, 2009) and tumorigenesis (Shen et al, 2007). The study of DNA methylation is of particular importance in cancer, as causal involvement has been demonstrated and it is the most stable of all epigenetic modifications, making it a desirable marker for both early detection and treatment of tumors. Hypermethylation of CpG sites in gene promoter regions leads to decreased gene expression; if such a gene is a tumor suppressor, this leads to carcinogenesis. To date, there have been no studies of genome-wide DNA methylation profiling of NETs. This study sets out to determine the DNA methylation profiles of low, intermediate and high grade pancreatic NET liver metastases with the intention of identifying dysregulated biological pathways in the development of these tumors. A protocol for the analysis formalin-fixed paraffin embedded tissue (FFPE) has also been developed in order to study these tumors in significant numbers following this pilot study.

#67 Interest of combined chromogranin A and pancreatic polypeptide for diagnosis and follow-up of gastroenteropancreatic endocrine carcinoma
Introduction: Assessment of tumor burden changes is essential for the management of well-differentiated gastroenteropancreatic neuroendocrine carcinoma (GEPNET). Chromogranin A (CgA) is the principal tumor marker for such tumors; however, its use to evaluate morphological tumor progression is not validated. Combined CgA and pancreatic polypeptide (PP) may increase sensitivity in the diagnosis of GEP-NET.
