#2739 Epigenetic Landscape of Pancreatic Neuro-Endocrine Tumors Reveals Distinct Cells of Origin and Means of Tumor Progression
Introduction: Pancreatic Neuroendocrine Tumors (PanNETs) arise from cells of the Islets of Langerhans. The majority of PanNETs are non-functional and their cell of origin cannot be defined analysing specific hormone production. Cell of origin assessment has proven importance to identify risk factors, prevent tumour development, and tailor treatment in many malignancies. Recent data on super-enhancer signatures has suggested a potential origin of PanNET from α- or β-cells. We and others have shown that distinct epigenetic profiles assessed by DNA methylation (DNAme) characterize genomic and prognostic groups of PanNET.

#477 Glucagon-Like Peptide 1 Receptor (GLP-1R) Imaging for the Preoperative Localization of Benign Insulinomas in 30 Patients
Introduction: Biochemical diagnosis of endogenous hyperinsulinemic hypoglycemia is straightforward. Surgical removal of an insulinoma is hampered by difficulties to localize it using conventional radiological procedures. In vitro data suggest that human insulinoma cells exhibit a high density of GLP-1R. 111In-exendin-4 is an 111In labeled GLP-1R agonist that binds with high affinity to GLP-1R and may be helpful in localizing insulinomas.

#975 Electron Microscopy of Pancreatic Beta Cell Neuroendocrine Tumors (NETs) in Multiple Endocrine Neoplasia Type 1 (MEN1) Knockout Mice Reveal an Adenomatous Phenotype with Depletion of Insulin Granules and Increased Mitochondrial Content
Introduction: Insulinomas are β cell neuroendocrine tumors (NETs) that secrete insulin, and ~4% of insulinoma patients have multiple endocrine neoplasia type 1 (MEN1). One in ten MEN1 patients present with an insulinoma. Previous reports describe insulinomas as possibly having typical granules (46.4%) or atypical smaller granules (34.3%), both associated with adenomas, or agranular cells (14.3%) associated with carcinomas. MEN1 knockout (Men1+/-) mice develop pancreatic NETs that are mostly insulinomas, and their ultrastructural phenotype is unknown. We hypothesized that these may resemble the adenoma phenotype with atypical granules.

#1850 Influence of VEGF Splicing on Microvessel Density and Architecture in Pancreatic Neuroendocrine Tumours
Introduction: Hypoxia in human tumours is associated with a poor prognosis and is characterized by the stabilization of HIF1α, upregulation of VEGF, and a high microvessel density (MVD). Interestingly, although pancreatic neuroendocrine tumours (pNETS) showing hypoxia signalling also have a poor prognosis, they paradoxically display a low MVD. We hypothesized that this paradoxon might be explained by alternative splicing of VEGF.
