#108 Everolimus (RAD001) induces tumor response in patients with medullary thyroid cancer: an in vivo and in vitro study
Introduction: Medullary thyroid cancer (MTC) is a neuroendocrine tumor (NET) of the thyroid C cells which is known to express somatostatin receptors. Octreotide, a somatostatin analogue exerting both antisecretory and antitumor activity in different types of NETs, has shown low efficacy in MTC. Everolimus (RAD001), an inhibitor of mTOR, has shown antitumor effects in patients with NETs and synergistic effects when combined with octreotide. Until recently, RAD001 has never been evaluated in MTC.

#1758 Efficacy and Safety of Low Dose Vandetanib in Metastatic Medullary Thyroid Cancer
Introduction: Medullary thyroid cancer (MTC) is an aggressive thyroid malignancy. Vandetanib is a tyrosine kinase inhibitor approved for locally advanced or metastatic MTC, administered orally 300 mg/daily. Diarrhea, hypertension and QT prolongation are common side effects and may require withdrawal or dose reduction.

#2156 Two Cases Synchronous Atypical Parathyroid Adenomas and Papillary Thyroid Carcinoma
Introduction: A literature review revealed a prevalence of approximately 3% of nonmedullary thyroid cancer, which was found in patients operated on for primary hyperparathyroidism (pHPT). Papillary thyroid carcinomas(PTC) is a malignant epithelial tumour. PTC represent up to 87% of all thyroid carcinomas. Atypical parathyroid adenoma(APA) are a subset of parathyroid neoplasms that exhibit some of the features of parathyroid carcinoma but lack unequivocal invasive growth. APA represents about 0,5-4% of cases of pHPT. As a group, they may be considered tumors of uncertain malignant potential.

Introduction: MTC is a well-differentiated neuroendocrine tumor in which somatostatin receptor (sst) expression is higher for sst1 and sst5 than for sst2. This may explain why the available sst2–selective analogues do not work in these patients and why pasireotide (SOM230), a novel, multi-receptor targeted somatostatin analogue with high-binding affinity for sst1, 2, 3 and sst5 could be effective.

#48 Preliminary report of the use of everolimus in a patient with progressive medullary thyroid carcinoma
Introduction: Medullary thyroid carcinoma (MTC) accounts for about 5-10% of all thyroid cancers, and while disease localized to the thyroid can be cured by surgery, there is no effective treatment for disseminated MTC. Constitutive activation of the RET proto-oncogene is implicated in familial and a proportion of sporadic cases, and up-regulates the AKT/PI3-kinase/mTOR signal transduction pathway. Recent studies have shown that the inhibitor of mTOR, everolimus (RAD001, Novartis, Basel), can inhibit the proliferation of the TT cell line and human MTC tumor cells in vitro. Clinically, everolimus has been shown to attenuate the progression of some gastrointestinal neuroendocrine tumors, but its clinical use in MTC has not yet been evaluated.
