#100 Mammalian target of rapamycin (mTOR) expression analysis and therapeutic approach in lung and gastroenteropancreatic poorly differentiated endocrine carcinoma (PDEC)
Introduction: PDEC clinically include gastroenteropancreatic PDEC, small cell lung cancer (SCLC) and large cell neuroendocrine carcinoma (LCNEC). These tumors are clinically similar and aggressive and they are usually treated with platinum compounds. mTOR signalling pathway has emerged as a promising target for well-differentiated endocrine carcinoma therapy. Because of varying behavior, PDEC are usually excluded from clinical trials employing the mTOR inhibitor RAD001.

Introduction: Grade 3 (G3) neuroendocrine neoplasms (NEN), arising from the gastro-entero-pancreatic system (GEP), are classified according to WHO 2010 and defined as having a Ki67 of > 20%. However this groups together G3 well differentiated neuroendocrine tumours (WD-NET) with poorly differentiated neuroendocrine carcinomas (PD-NEC). Introduction: Although much progress has been made in the past 2-3 years in terms of understanding signaling pathways in gastroenteropancreatic neuroendocrine tumors (GEP-NENs), approved treatment options are still limited. Cancer Immunotherapy has been announced as the breakthrough of the year in 2013 and might become an integral part of the clinical management strategy for solid tumors including GEP-NENs. Therefore, to develop immunotherapeutic strategies against NENs we need to know which immune escape mechanisms are relevant in high and low grade NEC/NENs.
