#2075 Phase 1 Pharmacokinetic and Pharmacodynamic Study of APOC, a New Controlled Release Formulation (CRF) of 15mg Octreotide Acetate in Healthy Male Volunteers
Introduction: Somatostatin analogues (SSAs) are considered the gold standard for systemic therapy of advanced neuroendocrine tumors (NETs). Octreotide is one of the SSAs most widely used in long-term therapies of NETs. There is increasing evidence that clinical benefits could be obtained with higher SSAs circulating levels but remain unreachable with current products without impacting significantly the quality of life of the patients. APOC is a new injectable controlled release technology containing Octreotide invented and developed by Ascil-Biopharm and designed to cover specifically these clinical unmet needs. It is presented as ready-to-use and can easily be manufactured at selected doses and durations in prefilled syringe.

#131 Efficacy and safety results from a Phase II study of pasireotide (SOM230) in the treatment of patients with metastatic NETs refractory or resistant to octreotide LAR
Introduction: Pasireotide, a multi-receptor targeted somatostatin analogue, has 30-, 5- and 39-fold greater affinity for sst1,3 and sst5 receptors, respectively, and a slightly lower affinity for sst2, than octreotide. Because of this multi-receptor binding profile, pasireotide may be effective in controlling symptoms of carcinoid syndrome in patients with gastroenteropancreatic neuroendocrine tumors (NETs) who are no longer responsive to currently available somatostatin analogues.

Introduction: Outcome measures reported by pts help to weigh benefit/effect of a therapy on quality of life (QoL), provided that their utility in clinical practice is proved. There are few QoL QNRs for pts with NETs, like QLQ-GINET21, with 21 items assessing muscle/bone pain, body image, information, sexual function, symptoms, social function and disease-related concerns.

#1725 Safety of Lanreotide 120 mg ATG (LAN) in Combination with Metformin (MET) in Patients (pts) with Progressive Advanced Well-Differentiated (WD) Gastro-Intestinal (GI) or Lung Carcinoids. A Pilot, One-Arm, Open-Label, Prospective Study: The MetNET-2 Trial
Introduction: Anti proliferative effect of LAN ATG 120 mg has been investigated in 204 GI-NET pts in Clarinet study with a 53% reduction in risk of death/progression vs placebo. An increasing number of trials have associated MET with better outcome in cancer pts due to a reduction of insulin/IGF1 levels and modulation of AMPK and TSC1-2/mTOR. The retrospective PRIME-net study conducted on 445 Italian pts, suggest that addition of MET to everolimus and/or SSA provides clinical benefit in diabetic pancreatic NETs.
