Characterization of Epigenetic Modulation in Pancreatic Neuroendocrine Neoplasms Abstract #2827

Introduction: Pancreatic neuroendocrine tumors (PanNETs) represent a rare and heterogeneous tumor entity. Despite advances in therapy cancer progression and resistance mechanisms are still challenging. Recent genome wide sequencing analyses in PanNETs identified a large number of mutated genes involved in epigenetic modulation.
Aim(s): Targeting epigenetic regulation in tumor cells in combination with specifically triggered immune cells could be a new therapeutic avenue.
Materials and methods: The human PanNET cell lines BON-1, QGP-1, NT-3 and NT-18 the murine insulinoma cell lines beta-TC-6 and RIN-T3 and the human monocyte-like THP-1 cells were polarized in M0/M1/M2 phenotypes and treated with Panobinostat (PB) and analyzed for functional effects and affected signaling pathways performing Western blot, FACS and qPCR analyses. Additionally, the impact of PB on microRNA and lncRNA expression in BON-1 and QGP-1 cells was investigated by RNA sequencing.

#2215 Development of Multiplex Biomarker Assay to Subtype Pancreatic Neuroendocrine Tumors (PanNETs) with Distinct Prognosis and Mutations
Introduction: Overall 5-year survival for PanNETs ranges from 25-100%. The treatment paradigm for PanNETs largely based upon grades, which is significantly heterogeneous. We previously defined three molecular subtypes (distributed grades) of PanNETs using PanNETassigner signature. There is an unmet clinical need for prognostic and predictive biomarkers and clinically-relevant assays to complement grade and improve patient stratification.
