Mammalian Target of Rapamycin Pathway Activation is Associated with RET Mutation Status in Medullary Thyroid Carcinoma

Introduction: The genetic pathways involved in medullary thyroid carcinoma (MTC), except for RET mutations, are largely unknown, as well as the detailed expression mapping of intracellular signalling transducers.

Aim(s): To screen for mutations in other genes downstream to RET pathway and to detect the activation patterns of mTOR signalling-related molecules.

Materials and methods: Sixty-five cases of MTC were selected for mutational analysis of RET, BRAF, N-, H- and KRAS, and PI3 kinase genes and for immunohistochemical analysis using antibodies against phospho-mTOR, phospho-p70S6K, phospho-AKT, Stat-3, phospho-MAP kinase and galectin-3.
