MEK/RAF and PARP as Novel Targets in Neuroendocrine Neoplasms – First Results from a Molecular in vitro Tumor Board Abstract #2974

Introduction: Neuroendocrine neoplasms (NEN) are a heterogeneous group comprising well differentiated neuroendocrine tumors (NET) and poorly differentiated neuroendocrine carcinomas (NEC). Although, major progress has been made in the development of targeted therapies in various cancers, establishment of a promising personalized therapy in NEN is still pending. So far, a lack of suitable preclinical models represent a major challenge concerning the validation of a mutational based targeted therapy (MBTT)
Aim(s): The present study investigates the opportunity of a MBTT in NEN cell lines.
Materials and methods: Using our newly established panel of 4 NET (NT-3, NT-18P, NT-18LM, NT-36) and 2 NEC (NT-32, NT-38) cell lines, a targeted exome sequencing of a 700 cancer gene panel was performed. After identification of potential therapeutic targets, cell lines were treated with specific inhibitors.

#38 Expression of mTOR signaling pathway and effects of mTOR inhibitors on its pathway in human neuroendocrine tumors
Introduction: Neuroendocrine tumors (NET), especially well-differentiated endocrine tumors, are often slow-growing, indolent, and may not become clinically apparent until the manifestations of metastatic spread or carcinoid syndrome. Results of previous studies demonstrated that phosphoinositide 3-kinase (PI3K)/Akt/mTOR signaling pathway may be activated in the great majority of NET cells. mTOR inhibitors are therefore considered effective anti-tumor reagents but they may also induce an activation of PI3K/Akt and MAPK pathways as a compensatory action and reduce their own anti-tumor activities.

#42 Chemotherapy in the Treatment of Progressive, Undifferentiated Neuroendocrine Carcinomas: a Single-Center Experience.
