The Anticancer Mechanism of Metformin in Pancreatic Neuroendocrine Tumor Cells

Introduction: Metformin, a widely used antidiabetic drug, has been reported to display potent anticancer properties in various types of cancers. In P-NETs, aryl hydrocarbon receptor-interacting protein (AIP) acts as a tumor suppressor and is up-regulated by the somatostatin analog octreotide.

Aim(s): We investigated the effects of metformin on P-NET cell proliferation, apoptosis and colony formation, the interaction of metformin and octreotide, and the involvement of the AIP pathway in these events.

Materials and methods: Brdu incorporation assay was used to investigate the effect of metformin on P-NET cell proliferation and its interaction with octreotide. Apoptosis was evaluated by flow cytometry and colony images by brightfield microscopy. The involvement of the AIP pathway in these events was evaluated by western blot and siRNA trasfection
