The Splicing Machinery Is Dysregulated in Low Grade Pulmonary Neuroendocrine Tumors Abstract #3053

Introduction: Alternative splicing dysregulation is increasingly regarded as a novel cancer hallmark influencing all key tumor features. We have shown that neuroendocrine tumors (NETs) overexpress aberrantly spliced variants of neuropeptides and receptors that increase tumor malignancy. More recently, we discovered that the splicing machinery is dysregulated in pituitary and pancreatic NETs. To date, the status of this machinery in low grade pulmonary NETs is largely unknown.
Aim(s): To assess the expression profile of a selected set of components of the splicing machinery and explore their clinical relevance in low grade pulmonary NETs.
Materials and methods: A custom-made microfluidic based qPCR array was used to measure the expression of 45 key components of the splicing machinery in 33 pulmonary NETs samples (including typical and atypical carcinoids) compared to their non-tumoral adjacent tissue, and 5 healthy lung samples. Possible associations between gene expression levels and relevant clinical parameters were assessed.

#111 Atypical lung carcinoid tumor metastatic to endocrine glands and bone marrow
Introduction: Bronchopulmonary neuroendocrine tumors (BP-NETs) comprise 20% of all lung cancers. Typical low grade carcinoid tumors (TC) represent the majority of BP-carcinoids. Intermediate grade atypical carcinoid tumors (AC) are less often encountered but more aggressive. Although metastatic dissemination to mediastinal lymph nodes, liver, skeleton and central nervous system (CNS) is common, involvement of the endocrine glands has rarely been reported.

#3056 Multi-Omic Characterization and Evolution of Neuroendocrine Neoplasm Organoids
Introduction: Molecular characterizations of neuroendocrine neoplasm (NENs) have unveiled candidate alterations associated with aggressiveness and suggested that the molecular link between neuroendocrine tumors (NETs) and neuroendocrine carcinomas (NECs) might be subtler than initially thought. For example, we have recently unveiled a new entity of pulmonary carcinoids (supra-carcinoids) with carcinoid-like morphology yet the molecular and clinical features of large cell neuroendocrine carcinoma (LCNEC). Testing hypotheses to explain aggressiveness and the possibility of progression or transition from NET to NEC through the accumulation of genetic anomalies requires in vitro and in vivo experimental models.

#2935 Prior Surgical Resection and Ga-68-DOTANOC PET/CT Imaging-Based Parameters: Do These Impact Outcomes in Grade II Gastroentero-Pancreatic Neuroendocrine Tumors (GEP-NETs)
Introduction: Grade 2 GEP-NETs have inherent tumor heterogeneity seen with positive expression of somatostatin (SSTR) and glucose (GLUT) receptors. Prior surgical resection and imaging parameters on Ga-68 DOTANOC PET/C are well-established variable impacting prognosis in well-differentiated grade 1 NETs. Since the degree of differentiation is variable in Grade 2 NETs, these parameters have not been studied.
