Detection of SSTR2 and 5 Expression on Circulating Tumour Cells in Neuroendocrine Tumours

Introduction: Neuroendocrine tumours (NET) overexpress somatostatin receptors (SSTR) which can be targeted for therapy with somatostatin analogues (SSA) or radionuclide treatment.

Aim(s): To develop an assay to detect SSTR2 and 5 expression on CTCs in NET patients.

Materials and methods: MCF7 cells were transfected with plasmid DNA carrying human SSTR2 or 5 and spiked into donor blood for testing using the CellSearch platform. Optimum antibody concentrations and exposure times were determined. Flow cytometry was used to assess whether marker detection was affected by presence of SSA. For clinical evaluation, 7.5mls patient blood was analysed by CellSearch and SSTR2/5 immunohistochemistry (IHC) was performed on matched FFPE samples.
