#4591 Efficacy of Trabectedin and Olaparib in homologous repair deficient neuroendocrine neoplasms – A subgroup analysis of the TOP-ART / PMO-1603 trial

Introduction: Genomic alterations resulting in homologous recombination deficiency (HRD) occur in a variety of cancers including neuroendocrine neoplasms (NEN). HRD-positive tumours are sensitive to PARP inhibitors such as olaparib. The DNA minor groove binder trabectedin leads to DNA double strand breaks and PARP activation. The combination of Trabectedin + Olaparib (TrO) may therefore have synergistic effects in HRD-positive tumours.

#3953 Olaparib use in a patient diagnosed with BRCA2 mutated adrenocortical carcinoma: A case report

Introduction: Adrenocortical carcinoma (ACC) is a highly lethal endocrine malignancy with an estimated annual incidence of 1.5–2 cases per million. Only a small percentage of ACC cases are associated with hereditary cancer syndromes while, equally only a few cases, have been reported with BRCA mutation. Targeted therapy of ACC with germline mutations is an area of research which renders the treatment approach challenging.

#3813 First report of the phase I LuPARP trial assessing 177Lu-DOTATATE in combination with the PARP inhibitor olaparib in patients with somatostatin positive tumors

Introduction: Patients with metastatic somatostatin receptor positive tumors can be treated with peptide receptor radionuclide therapy (PRRT) with 177Lu-DOTATATE. However, all patients do not respond sufficiently, and further treatment optimization is highly warranted. We hypothesize that the tumoricidal effect could be increased by adding a PARP inhibitor which will decrease the ability of the tumor cells to repair DNA-damage, making them more sensitive to radiation and increase the probability of tumor cell death.
