Development of Pre-Clinical NEN Models – Limitations and Opportunities

Introduction: Pre-clinical research in neuroendocrine neoplasia (NEN) has long been impaired by the paucity of NET and NEC cell lines. Furthermore, established models have not been widely available or displayed unusual features (eg. p53 and ras mutations, high proliferation rate).

Aim(s): In order to improve pre-clinical research opportunities in neuroendocrine neoplasia, we started a cell line development program in 2013.

Materials and methods: Fresh surgical samples were collected and subjected to mechanical and enzymatic digestion. Subsequently, a primary tumor cell culture was started in RPMI supplemented with 10% FCS and EGF/FGF2.
