#3799 High-dose somatostatin analogues’ effects on glycolipid metabolism in patients with neuroendocrine tumors

Introduction: Somatostatin analogues (SSAs), octreotide and lanreotide, represent the cornerstone medical therapy of both sporadic and hereditary G1-G2 neuroendocrine tumors (NETs). Changes in glucose metabolism have been shown to occur during standard-dose SSA therapy, but no changes during high-dose therapy have been evaluated.

#3764 Therapeutic response of ACTH-dependent ectopic Cushing's syndrome to low doses of osilodrostat

Introduction: Cushing's disease (CD) is the main cause of ACTH-dependent Cushing's syndrome (ACTH-CS). In 5–15% of cases, ACTH/CRH production is ectopic and is caused by neuroendocrine tumors of the lung, thymus, or pancreas, medullary thyroid carcinoma, and pheochromocytoma. Diagnostic imaging can locate the primary tumor; however, between 8-22% the cause is unknown. The FDA approved Osilodrostat for the treatment of inoperable or refractory CD, its use in high doses in patients with ectopic CS has been reported.

#3739 Local tumor control in patients with neuroendocrine neoplasms treated with stereotactic ablative radiotherapy (SBRT)

Introduction: Neuroendocrine neoplasms (NEN) represent a diverse group of malignancies. Anatomic origin, histology and biological behaviour vary extensively. The treatment armamentarium includes surgery (first choice when possible), systemic therapy and localised treatment modalities. The role of radiotherapy is not yet established, but studies indicate that local tumor control (LTC) can be achieved by high-dose radiotherapy. Stereotactic ablative radiotherapy (SBRT) is high doses of radiation delivered to a small target, with potential of achieving local tumor control.

#3145 Reduction of serum insulin levels and diazoxide doses as an effect of PRRT in patients with severe hypoglycemia due to primary non-operative insulinoma

Introduction: Severe hypoglycaemia in a course of inoperable insulinoma may be life-threating and it is not always well controlled even by high doses of diazoxide, which in some cases cause a significant toxicity. Nowadays, other forms of effective therapy are not available - use of protein kinase inhibitors (everolimus, sunitynib) sometimes bring satisfactory effect but is often associated with the risk of serious side effects. Use of Peptide receptor radionuclide therapy (PRRT), characterized by relatively low toxicity, is not widely used.

#2087 Prospective, Multi-Center, Open Label, Phase Study II of APOC, a New Controlled Release 15mg Octreotide Acetate, for the Treatment of Advanced NETs

Introduction: Somatostatin analogues (SSAs) are the cornerstone of systemic therapy of advanced NETs. The efficacy controlling hormone release and tumor growth together with the favorable toxicity profile have positioned SSAs as upfront therapy. Despite of second-line therapies including targeted agents, chemotherapy and radiolabeled peptides have demonstrated activity controlling tumor growth, the less favorable toxicity profiles become a usual discussion with patients (pts) to preserve the quality of life (QoL). Additionally, from concordant flow of evidences clinical benefits could be obtained with higher SSAs circulating levels but remain unreachable with current products. APOC is a new controlled release ready-to-use convenient therapeutic, designed to cover specifically these clinical unmet needs.
