Introduction: SSA is indicated for the treatment of G1 and some G2 gastroenteropancreatic neuroendocrine tumours (GEP-NETs) of midgut, pancreatic or unknown origin in adult patients with unresectable locally advanced or metastatic disease. As these types of cancer have much lower proliferation rates than common cancers, they are not exclusively considered cancer. It is hypothesised these patients may subsequently receive sub-standard cancer care. Furthermore, patients who are diagnosed with metastatic G3 neuroendocrine cancers will require systemic treatment with significant side effect profiles impacting negatively on quality of life.

Introduction: Peptide Receptor Radionuclide Therapy (PRRT) has shown positive clinical outcomes in somatostatin receptor (SSTR) overexpressing positive well-differentiated neuroendocrine tumors in combination with somatostatin analogue therapy. A noted decreased progression-free survivability in the use of PRRT has been noted in larger lesions (>3cm).

Introduction: Well-differentiated gastroenteropancreatic neuroendocrine tumors (GEP-NETs) are characterized by high-density expression of somatostatin receptors (SSTRs). RYZ101 (225Ac-DOTATATE) is a highly potent alpha-emitting radiopharmaceutical therapy (RPT) being developed for SSTR+ solid tumors. ACTION-1 is a 2-part, global, randomized, controlled, open-label, Phase 1b/3 trial of RYZ101. Part 1 (Phase 1b), an uncontrolled dose de-escalation study, is complete with no dose-limiting toxicities.
