#4364 Single agent axitinib in the management of patients with progressive pheochromocytoma and paraganglioma

Introduction: Radiolabelled therapy with meta-iodobenzguanine (MIBG) and chemotherapy with a combination of cyclophosphamide, vincristine, and dacarbazine can help manage patients with metastatic pheochromocytoma and paraganglioma (mPPGL). The phase II FIRSTMAPPP study supports the use of the tyrosine kinase inhibitor sunitinib in these malignancies, with median progression-free survival values of 8.9 versus 3.6 months and overall response rates of 31% versus 8% for sunitinib versus placebo. The median duration of sunitinib therapy was 11 months. Concurrent with this study, we launched a single-arm trial to examine the activity of a similar tyrosine kinase inhibitor, axitinib.

#4128 Validation of a 3-gene signature of response to axitinib in patients with advanced NETs

Introduction: Axitinib in combination with SSA significantly improved ORR and PFS per central blinded radiological assessment, as compared to placebo and SSA in 253 EP-NETs in the AXINET trial. The magnitude of the benefit, however, was limited. We recently described a 3-gene tumor transcriptomic signature predictive of response to axitinib in NET patients.

#3768 Predictive transcriptomic signature of response to axitinib in patients with advanced NETs included in the randomized, double-blind, placebo-controlled AXINET trial (GETNE 1107)

Introduction: Axitinib in combination with Octreotide LAR significantly improved ORR (13.2 vs 3.2%, OR 4.58, p=0.0045) and PFS (16.6 vs 9.9m, HR 0.71, p=0.017) per central blinded radiological assessment, as compared to placebo and SSA, in 256 patients with EP-NETs included in the AXINET trial (Garcia-Carbonero et al, ESMO 2021). The magnitude of the benefit, however, was limited.
