Activation of cdk4/Cyclind D1 and the Associated Attenuation of Rb Function in Pancreatic Neuroendocrine Tumors (Pan-NETs)

Introduction: Pan-NET poses challenges in clinical management due to the lack of identifications of key oncogenic pathways in this neoplasm. There are no reliable biomarkers to predict the recurrence and progression and treatment option is limited. Genetic changes in Rb pathway can lead to Pan-NET formation in mice. Genomic investigations in humans have not revealed Rb pathway mutation.

Aim(s): To investigate abnormalities in the Rb pathway in Pan-NETs.

Materials and methods: IHC was carried out on tissue microarray of Pan-NET. qRT-PCR was performed using mRNA from fresh samples of Pan-NETs. Gene maplification was performed by PCR and by FISH. Compound PD 0332991, a specific cdk4/6 inhibitor, was tested in Pan-NET cell lines.
