Immunohistochemical profiling of lung carcinoid subtypes and marker comparison of matched primary and metastatic tumors

Introduction: Multi-omic studies have identified three lung carcinoid (LC) subtypes (A1, A2, B) with unique gene expression for OTP, ASCL1 & HNF1a.

Aim(s): Here spatial expression differences for TTF1 and S100 were evaluated, in relation to subtyping of LC, by comparing primary tumor and matching metastasis.

Materials and methods: Surgically resected pathology revised LC (2003-2012) from the Dutch Pathology Registry and the Netherlands Cancer Registry (n=478) were evaluated. H-scores were determined for IHC staining of OTP, ASCL1, HNF1a, TTF1 and S100. TTF1 and S100 were considered positive if H-score > 0. S100 staining was classified as sustenticular, nuclear and cytoplasmic staining. Furthermore, 19 matched primary-metastatic samples were analyzed to investigate changes in Ki-67 proliferation index, H-score and subtype classification by Spearman’s correlation.
