MEN1 deficiency-induced the activation of β-Catenin-MGMT axis promotes Pancreatic Neuroendocrine Tumor growth and disrupts the response to Temozolomide

Introduction: O6-methylguanine DNA methyltransferase (MGMT) removes alkyl adducts from the guanine O6 position (O6-MG) and repairs DNA damage. High MGMT expression results in poor response to temozolomide (TMZ).

Aim(s): The biological importance of MGMT and the mechanism underlying its high expression in PanNETs remain elusive.

Materials and methods: By using pancreas-specific MEN1 KO mice and patient clinical tissue microarrays, we evaluate the association between MEN1 and MGMT expression, and further explore the underlying mechanism of MEN1 deficiency-regulated MGMT expression and TMZ tolerance of PanNETs both in vitro and in vivo.
