Protein validation of multi-omic classified pulmonary carcinoid – Identification of unique clinical subtypes and potential therapeutic vulnerability

Introduction: Multi-omic studies have identified three pulmonary carcinoid (PC) subtypes (A1, A2, B) with unique expression for OTP, ASCL1 & HNF1a.
Aim(s): To establish an immunohistochemical (IHC) panel that identifies PC subtypes and clinical relevance.
Materials and methods: All surgically resected PC (2003-2012) were identified from the Dutch cancer/pathology registry and pathology revised. IHC expression for OTP/ASCL1/HNF1a was scored in a blinded fashion for mRNA-profiled PC (n=5 per A1/A2/B) and PC cohort n=426. Distant normal lung tissue (≈35/type) was evaluated for neuroendocrine cell hyperplasia ((DIP)NECH). Finally, IHC expression for potential therapeutic targets SSRT2a & DLL3 was scored. All IHC was assessed by H-score.
