Introduction: Neuroendocrine carcinomas of the gastrointestinal tract (GEP-NEC) are rare but extremely aggressive tumor diseases with a 5-year survival rate of approximately 25%. The inactivating ARID1A mutation can be detected in approximately 40% of cases of GEP-NEC. Previous studies have shown that Aurora kinase A inhibition leads to selective synthetic lethality of ARID1A-deficient colorectal tumor cells.

#3852 Novel insights to study the potential of the somatostatin system in NETs and NECs
Introduction: Neuroendocrine tumors (NETs) and carcinomas (NECs) are a heterogeneous group of cancers with increasing incidence worldwide. Surgery is the only curative choice, whereas an effective pharmacological approach for disseminated disease is still lacking. Somatostatin (SS) receptors (SSTs) provide a suitable pharmacological target to treat NETs with SS analogues (SSAs). However, a large proportion of NETs and, particularly most NECs, are unresponsive or resistant to SSAs. The underlying causes remain poorly understood and alternative strategies are needed. Recently, cortistatin (CST), a neuropeptide analogue of SS with comparable affinity to SSTs, emerged as an anti-inflammatory player, with potential antitumoral features.

Introduction: Cancer-associated fibroblasts (CAF) have been identified as relevant contributors to cancer progression and drug resistance in many tumors. Although neuroendocrine tumors (NET) are often associated with a strong stromal reaction, no study has addressed whether CAF are involved in progression and therapeutic resistance in NET.

Introduction: Streptozotocin based chemotherapy is standard of care in patients with advanced pancreatic neuroendocrine tumors (pNET). Although partial remission can initially be achieved in up to 40% of patients, primary non-responders and development of secondary progression under therapy is a major clinical challenge. So far, the mechanism leading to streptozotocin resistance have not been elucidated.
