Introduction: Gastroenteropancreatic neuroendocrine tumors (GEP-NETs) are frequently diagnosed at advanced stage. For well-differentiated somatostatin receptor-positive (SSTR +) NETs, somatostatin analogues (SSAs) are the preferred first-line therapy. However, in patients (pts) with G2/G3 and a high tumor burden, SSA alone might not be enough.

Introduction: Non-functioning (NF), sporadic, G1-G2 pancreatic neuroendocrine tumors (PanNETs) usually display an indolent course. Surgery is the first-choice treatment for localized tumors >2 cm. Unresectable or metastatic PanNETs expressing somatostatin receptors (SSTRs) are treated with somatostatin analogues (SSAs). The PROMID and the CLARINET studies proved the SSA antiproliferative effect in advanced gastroenteropancreatic neuroendocrine tumors (GEP-NETs). The standard treatment for patients with PanNETs ≤2cm is active surveillance (AS). Yet no evidence of the value of SSA treatment exist in such patient population.

Introduction: Medullary thyroid carcinoma (MTC) is a well-differentiated neuroendocrine tumor of the thyroid and accounts for 1%-2% of thyroid cancers in the United States. MTCs present with locoregional metastasis in 50% of patients and distant metastasis in 10%-15% at diagnosis. Treatment options for recurrent or progressive disease according to RECIST 1.1 criteria, are limited. Tyrosine kinase-inhibitors (TKI) are preferred as first-line systemic therapy. Although somatostatin analogues (SSA) may play a role in MTC, limited evidence is available.
