#4670 The efficacy of first-line combination therapy with everolimus plus lanreotide for gastroenteropancreatic neuroendocrine tumour with a poor prognostic factor: Updated and subgroup analysis of the phase III study, STARTER-NET (JCOG1901)

Introduction: Everolimus monotherapy provided a limited median progression-free survival (PFS) for gastroenteropancreatic neuroendocrine tumours (GEP-NETs), there remains a necessity for better therapeutic approaches.

#4663 CAREFNDR: A phase III, randomised, parallel group, placebo-controlled study to evaluate the efficacy and safety of paltusotine in adults with carcinoid syndrome due to well-differentiated neuroendocrine tumours

Introduction: Paltusotine is a once-daily, oral, nonpeptide, selective SST2 receptor agonist in development for carcinoid syndrome (CS) treatment. In a Phase 2, open-label, dose-ranging study, paltusotine reduced the frequency and severity of CS symptoms and was well tolerated (NCT05361668).

#4518 Efficacy and safety of cabozantinib (CABO) for advanced Grade 3 (G3) neuroendocrine tumours (NETs) after progression on prior therapy: Subgroup analysis of the phase III CABINET trial (Alliance A021602)

Introduction: Well-differentiated G3 NETs have been described as a distinct category recently, and randomised data regarding efficacy of therapy are scarce. In the phase 3 CABINET trial (NCT03375320), CABO significantly prolonged progression-free survival (PFS) compared with placebo (PB) in patients (pts) with advanced, previously treated, progressive well-differentiated extra-pancreatic NETs (epNETs) and pancreatic NETs (pNETs) of all grades (Chan et al., NEJM, 2024).

Introduction: Our multi-centre, prospective, open-label, two-arm parallel group, randomised, controlled, phase III study will address the efficacy and safety of adjuvant peptide receptor radionuclide therapy after curative surgery of locoregional limited small intestine neuroendocrine neoplasia.

#4380 Cell-free DNA concentration correlates with copy number variant (CNV)-count, describes tumour progression and nuclear instability in GEP-NETs

Introduction: Cell-free DNA (cfDNA) levels depend on various factors related to the shedding of cells and might provide valuable information on tumour state and treatment success. In a genomic profiling pilot study conducted as part of the COMPOSE Phase III multicentre open-labelled clinical trial we assessed multiple factors and their impact on cfDNA levels in 14 GEP-NET patients.
