Evidence for the role of miR196a in regulating small intestinal neuroendocrine tumor proliferation and metastasis via HOX/AKT pathway activation

Introduction: MicroRNAs (miRNAs) regulate cell proliferation, differentiation, and apoptosis functioning as tumor suppressor genes but their role in NET proliferation and metastasis has not been fully evaluated.
Aim(s): To use global approaches to identify miRNAs, their mRNA targets and functional signaling pathways up-regulated in malignant small intestinal NETs.
Materials and methods: An Affymetrix approach identified miRNAs in seven normal mucosa, four localized SINETs and seven hepatic metastases and four NET cell lines. Real-time PCR confirmed miRNA levels. For physiological studies, miR196a was knocked in cell lines using miRNA antisense. Proliferation, target transcript alterations (HOXB6,B7,C8) and AKT activation was determined.
