Tumor Microenvironment in Merkel Cell Carcinoma: Association with Merkel Cell Polyomavirus and Clinico-Pathological Features in a Retrospective Cohort Study

Introduction: MCC is a rare and aggressive skin cancer with a neuroendocrine phenotype.Association between MCC and Polyomavirus is known.PD-1 and PD-L1 have recently become key players in the therapeutic landscape.

Aim(s): To investigate: tumor microenvironment and its interaction with clinico-pathological features and outcomes;PD-L1 in cancer cells and in tumor-infiltrating lymphocytes (TILs) and its prognostic role;to assess viral status and its association with immune response and clinical outcomes.

Materials and methods: 72 pts affected by MCC were retrospectively analyzed;formalin-fixed,paraffin-embedded tissue samples were obtained from the Pathology files of the Città della Salute e della Scienza Hospital.All cases were reviewed and staged according to the current classification schemes.In a subgroup of 34,we performed IHC profiling for CD3, CD8, FoxP3, CD56,PD-L1.MCXPyV status was assessed with the CM2B4 clone.TILs were categorized as brisk, nonbrisk, absent.Statistical analysis were performed with Statistical Package for Social Science.
