#3961 Enhancer heterogeneity of lung carcinoids reveals sensitivity to FGF signaling inhibition
Introduction: Well-differentiated low grade lung carcinoids are histo-pathologically classified as typical and atypical, but each subtype is still heterogenous both at the molecular level and its clinical manifestation. Therefore, the treatment of these patients is challenging. The two main challenges are the lack of effective drug treatments and the lack of reliable biomarkers to guide management since the disease in patients with the same tumor grade/stage often has different clinical courses. The genetic, epigenetic, and developmental programs that drive lung carcinoids remain obscure, limiting our abilities to suggest new biomarkers and drug targets.

Introduction: NENs are treated with systemic therapies with limited non-curable efficacy due to development of drug resistance (DR). While cannabis and its extracts (cannabinoids, CE) are mostly used for their palliation properties, increasing evidence suggests an anticancer trait via several cellular pathways. Limited data exist on cannabis and the endocannabinoid system (ECS) in NENs and their impact on the development of DR.

Introduction: Patients with unresectable NENs are offered a variety of non-curable therapeutic options, which eventually fail due to drug resistance (DR). Increasing evidence suggest an anticancer trait of cannabinoids, via cellular pathways including mTOR, known to be associated with DR development. Still, limited data exist on the anti-cancer effects of cannabinoids in NENs.

Introduction: mTOR inhibitors (mTORi) such as RAD001 demonstrated promising anti-cancer effect in NETs. Autophagy, a cell survival mechanism, is activated by mTORi. We have recently shown in the human NET cell line BON1 that autophagy is essential for cell survival. Treatment with CQ alone or together with mTORi robustly inhibited cell proliferation and survival, suggesting that treatment with CQ may potentiate the anti-tumorigenic effects of mTORi.
