#4546 Epigenetic exploration of mesenteric fibrosis indicates epigenetic disruption of DAXX is driving metastatic potential in small intestinal neuroendocrine tumours

Introduction: Up to 50% of patients diagnosed with small intestinal neuroendocrine tumours (SI-NETs) are affected by mesenteric fibrosis (MF) which has significant pathological effects and impacts patient mortality.

Authors: Webster A, Martins M, Hodgetts H, Feelders R, Hofland L,

Keywords: DNA methylation, SI-NET, Mesenteric fibrosis, Epigenetics, Gene regulation, DAXX,

#4516 A methylation-based classifier for neuroendocrine tumour detection using smMIP technology

Introduction: DNA methylation plays an important role in NET pathogenesis. Distinct methylation profiles can differentiate between tumour subtypes, predict malignancy potential, and provide insights into tumour progression.

Keywords: neuroendocrine tumour, DNA methylation, biomarker selection, IMPRESS technology, classifier model,

#4505 NET-IMPRESS: A novel methylation-based assay to diagnose and monitor NET patients using liquid biopsies

Introduction: Previously, we showed that analysing copy number alterations (CNAs) in cell-free DNA (cfDNA) of NET patients using shallow whole genome sequencing (sWGS) is a potential biomarker for diagnosis and follow-up (PMID: 34759042). We now present NET-IMPRESS, an easy, cost-effective methylation-based assay for detecting circulating tumour DNA (ctDNA) in NET samples.

#4256 Epigenetic prediction of aging and metabolic traits provides insight into tumor biology in multi-focal ileal neuroendocrine tumors

Introduction: Approximately half of small intestinal neuroendocrine tumors (siNETs) are multifocal, and recent investigations into the genetic architecture of these tumors have found that they are not clonal. Due to the low mutational rate and slow growing nature of these small tumors, the ‘timing’ of the development of individual tumors has not been possible.

#4244 Cell-free DNA (cfDNA) methylation profiling for minimally invasive cancer detection in patients with Extra-Pulmonary NeuroEndocrine Carcinoma (EP-NEC)

Introduction: Robust biomarkers to aid clinical management of patients (pts) with EP-NEC are lacking. cfDNA methylation profiling has shown higher sensitivity/specificity than cfDNA genome profiling in diagnosing and classifying various cancers, including Small Cell Lung Cancer (SCLC).
