Introduction: SI-NETs are often associated with mesenteric fibrosis (MF) which causes significant morbidity and mortality. Telotristat ethyl (TE) was developed to treat carcinoid syndrome in SI-NET patients however, recent studies indicated TE could also have anti-tumor activity, the mechanism for which remains unclear.

Introduction: Carcinoid syndrome occurs in 20% patients, presenting with flushing, abdominal pain, diarrhoea, and wheeze. The standard of care for carcinoid syndrome is somatostatin analogues (SSAs) with add-on Creon, codeine and loperamide therapy. Nonetheless, half of patients experience debilitating diarrhoea. Telotristat-ethyl is a peripheral tryptophan-hydroxylase inhibitor approved for treatment of diarrhoea, supported by Phase-3 clinical trials.

#3290 Intratumoral serotonin might decrease the efficacy of PRRT in ileal neuroendocrine tumors
Introduction: Partial remission can be achieved by peptide receptor radionuclide therapy (PRRT) in only roughly 20% of patients with midgut neuroendocrine tumor (NET). This low remission rate can be due to the radioprotective role of stored intracytoplasmic serotonin in NET cells. If blocked, e.g. by telotristatethyl, PRRT efficacy might be increased.

Introduction: Clinical trials have demonstrated the efficacy and safety of telotristat ethyl (TE) treatment in combination with somatostatin analogues (SSAs) in patients with neuroendocrine tumors (NETs) and inadequately controlled carcinoid syndrome (CS) diarrhea. The TELEACE study was designed to evaluate the real-world effectiveness of TE in US clinical practice.
