#4364 Single agent axitinib in the management of patients with progressive pheochromocytoma and paraganglioma

Introduction: Radiolabelled therapy with meta-iodobenzguanine (MIBG) and chemotherapy with a combination of cyclophosphamide, vincristine, and dacarbazine can help manage patients with metastatic pheochromocytoma and paraganglioma (mPPGL). The phase II FIRSTMAPPP study supports the use of the tyrosine kinase inhibitor sunitinib in these malignancies, with median progression-free survival values of 8.9 versus 3.6 months and overall response rates of 31% versus 8% for sunitinib versus placebo. The median duration of sunitinib therapy was 11 months. Concurrent with this study, we launched a single-arm trial to examine the activity of a similar tyrosine kinase inhibitor, axitinib.

#4146 Anticancer activity of Cabozantinib and Temozolomide in cell lines derived from lung carcinoid and pancreatic neuroendocrine tumors

Introduction: Limited therapeutic options are available for patients with advanced neuroendocrine neoplasms (NENs). Cabozantinib (CBZ), a tyrosine kinase inhibitor including c-MET pathway, and Temozolomide (TMZ), an alkylating agent enhancing the anti-angiogenic effect of CBZ, are emerging as promising treatments in NENs.

#4128 Validation of a 3-gene signature of response to axitinib in patients with advanced NETs

Introduction: Axitinib in combination with SSA significantly improved ORR and PFS per central blinded radiological assessment, as compared to placebo and SSA in 253 EP-NETs in the AXINET trial. The magnitude of the benefit, however, was limited. We recently described a 3-gene tumor transcriptomic signature predictive of response to axitinib in NET patients.

Introduction: Surufatinib (SUR), an oral tyrosine kinase inhibitor, is a standard treatment for advanced NETs. Transarterial embolization (TAE) is a key local treatment for inoperable liver metastases from NETs.

#3795 Sex differences on multikinase inhibitors toxicity in patients with advanced gastroenteropancreatic neuroendocrine tumors – A pooled analysis of five clinical trials

Introduction: There is an increasing interest in the role of sex and gender in cancer patients. The impact of sex differences in systemic oncological therapies is still unknown, and there is a lack of evidence, especially in uncommon neoplasms like neuroendocrine tumors (NET).
