#4424 Tumour-derived apolipoprotein E induces tip endothelial cell to remodel tumour stroma ratio and promote pancreatic neuroendocrine tumour progression

Introduction: Current clinical trials have shown that single-targeted matrix administration does not improve patient prognosis. The investigation of targeted matrix administration in combination with other therapies is crucial for effectively managing TSR in pNETs.

#4319 Everolimus combined with orlistat inhibits the development of neuroendocrine tumours by inhibiting the mTOR-PI3K-Akt signalling pathway

Introduction: Everolimus, an mTOR inhibitor, has demonstrated efficacy in NET treatment. Orlistat, used primarily for obesity management, inhibits fat absorption. This study investigates the potential synergistic effects of Everolimus combined with Orlistat on the development of NETs.

#4316 Combination of sorafenib and everolimus regresses pancreatic neuroendocrine tumour significantly in vitro and vivo

Introduction: PNET is a rare group of highly heterogeneous tumours with neuroendocrine differentiation properties. Everolimus is the most promising drug for progressive well-differentiated pNETs. Sorafenib's anti-tumour activity may involve regulation of the mTOR pathway.

#4306 EVENET: Randomised phase II trial of everolimus 5mg vs. 10mg daily in patients with neuroendocrine tumours

Introduction: Everolimus 10mg PO daily is approved for patients (pts) with advanced G1/G2 neuroendocrine tumours (NET) but is associated with significant toxicity, including 20% of serious infections in real-world pts. In phase I trials, 5mg/day was sufficient to inhibit the mTOR pathway. Retrospective data suggest that 5mg/day is similarly effective to 10mg/day but less toxic.

#4304 Co-targeting the Endocannabinoid System (ES) and mTOR in NEN potentiates their anti-tumour effect via unique metabolic changes

Introduction: The current treatment of neuroendocrine neoplasms (NEN) has limited efficacy, highlighting the need for novel therapeutic approaches. We have previously shown the potential therapeutic benefit when combining ES blocking with Everolimus (Eve).
