#4506 Matching adjusted indirect comparison (MAIC) of [177Lu]Lu-DOTA-TATE (177Lu-DOTATATE) vs. streptozocin-based chemotherapies (STZ-BC) as first-line (1L) treatment for advanced grade 2/3 (G2/G3) pancreatic NETs (pNETs)

Introduction: The NETTER-2 trial showed improved progression-free survival (PFS) with 1L 177Lu-DOTATATE vs. high dose octreotide in advanced G2/G3 pNETs. No published evidence exists comparing 177Lu-DOTATATE with other recommended 1L treatments, such as STZ-BC, hence indirect comparisons need to be conducted.

Introduction: Aim of this study: The chemotherapy of metastatic pancreatic neuroendocrine tumors (TNEm) of low grade (G1, G2 Ki-67< 20%) is based on old studies and relies on streptozocin, the tolerance of which is poor. #3555 Risk of myelodysplastic syndrome/acute leukemia with sequential capecitabine/temozolomide and 177Lu-dotatate

Introduction: The treatment landscape for neuroendocrine tumors includes alkylating-agent chemotherapy and peptide receptor radiotherapy (PRRT) with 177Lu-Dotatate. The risk of MDS/AML associated with 177Lu-Dotatate is approximately 2-3%. Several small prior studies have suggested substantially higher rates of MDS/AML (approximately 10%) in patients who have also received alkylating agent chemotherapy with streptozocin or temozolomide, either combined with PRRT or sequentially.

#3403 Trends in chemotherapy use in neuroendocrine neoplasms: One step beyond guidelines

Introduction: The therapeutic landscape of neuroendocrine neoplasms (NENs) is constantly evolving. Current guidelines reflect the complexity of treatment sequences and mainly incorporate evidence from phase 3 clinical trials. Chemotherapy (CT) is a valuable option for patients with lung, pancreatic, and high-grade NENs, but proper phase 3 randomized studies are an unmet need. Nevertheless, several phase 2 studies with CT have been presented and published in the last year, with impact in both clinical practice and guidelines.
