#4601 Pembrolizumab combined with cisplatin or carboplatin and etoposide in treatment naïve advanced Merkel cell carcinoma (MCC): The phase II PANDORA Trial

Introduction: MCC is a rare and aggressive skin neuroendocrine tumour with high metastatic potential and mortality. Therapeutic options in metastatic/unresectable MCC are based on immunotherapy or platinum-based chemotherapy. However, a subgroup of patients (pts) has primary immunoresistance disease, so it would be important discover active upfront combinations. PANDORA trial [NCT 06086288] is an open-label, multicentre, single-arm phase II trial evaluating the activity and safety of pembrolizumab (PEM) combined with platinum-based chemotherapy as 1st line treatment in pts with metastatic, unresectable or recurrence MCC. #4537 The usefulness of circulating fibrosis markers in detection of carcinoid heart disease

Introduction: Carcinoid heart disease (CHD) is a potential serious fibrotic complication in patients with neuroendocrine neoplasms (NENs), affecting the heart valves. Early detection is crucial to avoid severe manifestations. NT-proBNP is suggested as an initial screening tool in detection of CHD; however, the value of NT-proBNP as a screening tool is questionable.

#4534 Development of new 3D in vitro models for pancreatic neuroendocrine tumours (PanNETs)

Introduction: Current PanNET in vitro models struggle to accurately replicate the tumour’s biology and microenvironment, limiting understanding of the mechanisms underlying PanNET behaviour and drugs response.

#4533 Receptor radionuclide therapy with 177Lu-DOTATOC (177Luedotreotide or 177Lu-octreotide) in SSTR positive patients: A multicentre, prospective, phase II trial

Introduction: For advanced G1-G2 GEP-NET patients, peptide receptor radionuclide therapy (PRRT) with 177Lu-DOTATATE can be recommended. However, there are several SSTR-positive tumours for which PRRT has previously shown efficacy that are not currently covered by the indications for 177Lu-DOTATATE in Italy and, therefore, cannot benefit from it. Less used and studied is the 177Lu-DOTATOC. This is a radiopeptide with excellent safety and efficacy profile in PRRT and certainly not lower than 177Lu-DOTATATE.

#4389 The LOLA trial: Phase II trial with combination of cabozantinib plus lanreotide in gastroenteropancreatic (GEP) and thoracic neuroendocrine tumours (NETs) – Interim results of step 1

Introduction: LOLA is an Italian, multicentre, open-label, double cohort, non-randomised, 3-stage, phase 2 trial aiming to assess the safety and activity of the combination of cabozantinib (CABO) + lanreotide (LAN) in patients (pts) with advanced or metastatic GEP and unknown primary NETs with Ki-67 > 10% or thoracic NET. In NETs sunitinib is the only TKI approved in advanced pancreatic NETs. While CABO has been reported superior to sunitinib in renal cancer its role in NETs is still investigational.
