#4654 Usefulness of soluble tumour necrosis factor-related apoptosis-inducing ligand (TRAIL) and its receptors R2 and R3 in progression assessment of gastroenteropancreatic neuroendocrine neoplasms – A preliminary study

Introduction: TRAIL is a member of TNF family and appears in membrane and soluble (s) forms. It acts either triggering (R1 and R2) or blocking (R3 and R4) apoptosis. TRAIL targeted therapy is widely studied and known as potentially effective in number of neoplasms.

#4607 BH3-mimetic drugs elicit cell death of neuroendocrine tumours in preclinical models – An emerging therapeutic strategy for NETs

Introduction: There is an unmet clinical need to identify new, effective therapies for patients with neuroendocrine tumours (NETs). Analysis of single-cell expression data revealed that NETs express high levels of the Bcl2 family of antiapoptotic proteins. Therefore, we hypothesised that proapoptotic drugs, such as BH3-mimetics, can induce programmed cell death, i.e., apoptosis, of neuroendocrine cancer cells.

#4524 SIRT7 drives the radioresistance of pancreatic neuroendocrine tumours via the DNMT1-MEN1 axis

Introduction: Pancreatic neuroendocrine tumours (PanNETs) are a rare and highly heterogeneous type of tumour in the pancreas. After failure of standard treatment, patients have poor prognoses. Radiotherapy may be a potential therapeutic modality for such patients. However, PanNETs usually exhibit a radiation “cold” tumour through unclarified mechanisms.

#4415 MGMT-activated DUB3 stabilises Nrf2/NCOA4 to modulate apoptosis and ferroptosis, driving temozolomide resistance in pancreatic neuroendocrine tumours

Introduction: O6-methylguanine DNA methyltransferase (MGMT) is a key DNA repair enzyme that removes methyl groups from O(6)-alkylguanine in DNA, introduced by temozolomide (TMZ). Clinical studies in pancreatic neuroendocrine tumours (PanNETs) have confirmed its role in TMZ efficacy.

#4372 CVM-005: A multicentre, single-arm, phase IIa study of CVM-1118, a novel oral anti-vasculogenic mimicry (VM) agent, in patients with treatment-refractory advanced neuroendocrine tumours (NET)

Introduction: NETs are highly vascularised tumours and a viable target for anti-VM therapies. CVM-1118 is an anti-tumour agent with multiple MOAs, including disruption of VM networks and induction of apoptosis via targeting mitochondrial TRAP1 protein.
