#4373 Focal adhesion kinase drives proliferation and invasion in gastrointestinal neuroendocrine tumours via modulation of transformative cell signalling and gene expression

Introduction: Focal Adhesion Kinase (FAK) is a non-receptor protein kinase that localises in both the cytoplasm and nucleus, influencing cell function through its enzymatic and scaffold activities. Through its scaffold function, FAK modulates gene expression epigenetically. Gastrointestinal neuroendocrine tumours (GI-NETs) exhibit a relatively low mutation rate, supporting the hypothesis that these malignancies may be driven epigenetically. Recently, PROTAC (PROteolysis TArgeting Chimeras) technology has enabled selective inhibition and degradation of FAK, providing a novel approach to explore its role in GI-NETs.

#4319 Everolimus combined with orlistat inhibits the development of neuroendocrine tumours by inhibiting the mTOR-PI3K-Akt signalling pathway

Introduction: Everolimus, an mTOR inhibitor, has demonstrated efficacy in NET treatment. Orlistat, used primarily for obesity management, inhibits fat absorption. This study investigates the potential synergistic effects of Everolimus combined with Orlistat on the development of NETs.

Introduction: The current treatment of neuroendocrine neoplasms (NEN) has limited efficacy, highlighting the need for novel therapeutic approaches. We have previously shown the potential therapeutic benefit when combining ES blocking with Everolimus (Eve).

#4150 Uncovering the role of netrins and DCC (deleted in colorectal cancer) in pancreatic neuroendocrine neoplasms (PNEN) tumorigenesis

Introduction: DCC functions as a tumor suppressor and is altered in various tumors, including neuroendocrine neoplasms. Netrin (NTN)-1 serves as the primary ligand for DCC. Operating as a dependence receptor, DCC induces apoptosis without NTN and promotes cell survival in its presence. In some cancers like small cell lung cancer and neuroblastoma, upregulation of NTN-3 rather than NTN-1 has been observed. However, the precise involvement of NTNs and DCC in PNEN remains unclear.

#4146 Anticancer activity of Cabozantinib and Temozolomide in cell lines derived from lung carcinoid and pancreatic neuroendocrine tumors

Introduction: Limited therapeutic options are available for patients with advanced neuroendocrine neoplasms (NENs). Cabozantinib (CBZ), a tyrosine kinase inhibitor including c-MET pathway, and Temozolomide (TMZ), an alkylating agent enhancing the anti-angiogenic effect of CBZ, are emerging as promising treatments in NENs.
