Introduction: Pancreatic neuroendocrine tumours (PanNETs) are classified into grades (G1-G3) based on mitotic rate, with higher grades exhibiting increased dedifferentiation and invasiveness. Despite the clinical significance of PanNETs progression, the underlying mechanisms remain largely unexplored. We have previously reported that altered alternative splicing plays an essential role in PanNETs progression, where SNW1, a key regulator of splicing and transcription, was found to be overexpressed in tumour tissue, correlating with higher grade and recurrence risk.

Introduction: Post-transcriptional modifications play crucial roles in regulating gene expression and cellular function. While recent findings indicate that alternative splicing dysregulation plays a key role in tumorigenic properties of neuroendocrine tumours (NETs), the role of other RNA processing mechanisms, such as N6-methyladenosine (m6A) — being the most common RNA methylation — remains largely unexplored in NETs.

#4208 The somatostatin system – A silent messenger in pheochromocytomas and paragangliomas?
Introduction: Pheochromocytomas and paragangliomas (PPGLs) are rare neuroendocrine neoplasms (NENs) arising from neural crest-derived chromaffin cells. Due to the abundance of somatostatin receptors (SSTs) on the surface of most NENs, somatostatin analogues (SSAs) have become a cornerstone in their diagnosis and treatment. However, while PPGLs exhibit this feature, their functional responsiveness to SSAs remains limited, with the underlying cause of this resistance being still elusive.

Introduction: Pancreatic Neuroendocrine Tumors (PanNETs) are characterized by a low number of mutations. Despite genomics, transcriptomics and epigenomics studies have helped to understand the molecular features of PanNETs, there is still a vast unexplored ground for better comprehension of this disease. In this context, we have previously documented that RNA splicing is dysregulated in these tumors, which unveils new avenues to discover potential biomarkers and therapeutic targets. However, clinical and molecular implications of this dysregulation are still very poorly understood.

#3859 Molecular landscape of splicing dysregulation in pheochromocytomas and paragangliomas
Introduction: Pheochromocytomas and paragangliomas (PPGL) are commonly benign neuroendocrine tumors (NETs). However, up to 25 % of patients develop metastases or aggressive behaviour. Genomic characterization current classifies PPGL into different genetic clusters, yet there are no biomarkers which could stratify patients based on their prognosis. The dysregulation of the splicing process has emerged as a novel feature, common in most cancers, and linked to a more aggressive phenotype in several tumors, including NETs, but this process has not been examined in detail in PPGL.
