Introduction: Gastrointestinal neuroendocrine tumors (GI-NETs) exhibit a complex and diverse nature, posing challenges in their effective treatment and management. While there have been advancements in treatment options, addressing the need for more effective and targeted therapies remains a significant issue, especially for patients with advanced or metastatic disease. Improving outcomes in the management of patients with GI-NETs is a key priority, and further research and development are crucial in this regard.

#3536 Comparison of drug sensitivity profiles of various NEN spheroids

Introduction: Neuroendocrine neoplasms (NENs) are rare cancers that arise from neuroendocrine cells. NENs are classified as well-differentiated neuroendocrine tumors (NETs) and poorly differentiated neuroendocrine carcinomas (NECs). Small bowel and pancreatic NETs (SBNET & PNETs) are generally slow-growing but commonly metastasize to the liver and become aggressive cancers. Little is known about the drug sensitivity profile of these 3 NENs due to the lack of cellular and animal models of these malignancies. Recently, we have developed a protocol to culture NEN cells from patient tumors as patient-derived spheroids (PDS) and showed expression of NEN markers. The rarity and slow growing features of these spheroids have been hindering drug screens with many compounds.

#3398 Patient-derived tumoroids phenocopy original gastro-entero-pancreatic neuroendocrine neoplasms and facilitate in vitro drug screening

Introduction: GastroEnteroPancreatic Neuroendocrine Neoplasms (GEP-NENs) are rare cancers that appear clinically and biologically extremely heterogenous. While highly proliferative cases are invariably lethal, slower-proliferating NENs—representing predominant cases—have unpredictable clinical courses varying from indolent to malignant. Recent comprehensive NGS screens in patients helped to extend our understanding. However, complementary relevant preclinical models to address disease heterogeneity and to investigate patient-specific treatment alternatives are still inexistent.
