Introduction: Neuroendocrine tumours (NETs) are a diverse group of neoplasms arising from neuroendocrine cells. Within this group, lung neuroendocrine tumours (LungNETs) represent one of the most frequent subtypes of NETs. LungNETs are classified by grade, according to their proliferative index and aggressiveness. Among them, typical carcinoids (G1) and atypical carcinoids (G2) are the lowest grade subtypes. While these tumours are less aggressive, they remain underexplored compared to higher-grade subtypes, highlighting the need for molecular studies to improve clinical approaches. In this context, previous studies have shown that abnormalities in the splicing machinery are frequent in tumour pathologies and also occur in LungNETs.

#4095 CAPTEM and PRRT to improve overall response for pancreatic NETs (pNETs) – The CORONET trial
Introduction: Patients with advanced pNETs have limited treatment options. The NETTER-2 and OCLURANDOM trials have established peptide receptor radionuclide therapy (PRRT) as a standard treatment in pNETs; CAPTEM chemotherapy is an active regimen in NENs. The CONTROL NETs trial has shown promising response rate and progression-free survival with PRRT+CAPTEM, warranting further investigation.

Introduction: Axitinib in combination with Octreotide LAR significantly improved ORR (13.2 vs 3.2%, OR 4.58, p=0.0045) and PFS (16.6 vs 9.9m, HR 0.71, p=0.017) per central blinded radiological assessment, as compared to placebo and SSA, in 256 patients with EP-NETs included in the AXINET trial (Garcia-Carbonero et al, ESMO 2021). The magnitude of the benefit, however, was limited.

Introduction: GastroEnteroPancreatic Neuroendocrine Neoplasms (GEP-NENs) are rare cancers that appear clinically and biologically extremely heterogenous. While highly proliferative cases are invariably lethal, slower-proliferating NENs—representing predominant cases—have unpredictable clinical courses varying from indolent to malignant. Recent comprehensive NGS screens in patients helped to extend our understanding. However, complementary relevant preclinical models to address disease heterogeneity and to investigate patient-specific treatment alternatives are still inexistent.
