Introduction: High-grade pancreatic neuroendocrine carcinoma (panNEC) pose challenges in terms of classification and treatment. While molecular genetics have made strides in deciphering significant inter-tumor heterogeneity, the extent of intra-tumoral heterogeneity and lineage plasticity remains unexplored.

#3804 Wide molecular analysis of PanNET revealed that only MGMT promoter methylation seemed to correlate with Temozolomide response

Introduction: Temozolomide (TMZ) is an alkylating agent and is widely used in the treatment of PanNET. So far, only the MGMT status, defined by its immunohistochemical (IHC) expression or its promoter methylation, has been proposed as predictor of response to TMZ, but its value remains controversial.

#3388 Epigenetic progression steps in alpha-lineage MEN1-DAXX/ATRX mutated Pancreatic Neuroendocrine Tumors (PanNET)

Introduction: Recently, based on DNA methylation profiles, we have hypothesized that PanNETs with mutations in ATRX, DAXX and MEN1 originate from α-cells. Alpha-like PanNETs, small and indolent, were enriched for mutations in MEN1 only, while epigenetically intermediate PanNETs, larger and with high relapse risk, showed mutations in both MEN1 and DAXX/ATRX (Int-ADM). Int-ADM PanNETs show progressive loss of alpha cell differentiation.

#1850 Influence of VEGF Splicing on Microvessel Density and Architecture in Pancreatic Neuroendocrine Tumours

Introduction: Hypoxia in human tumours is associated with a poor prognosis and is characterized by the stabilization of HIF1α, upregulation of VEGF, and a high microvessel density (MVD). Interestingly, although pancreatic neuroendocrine tumours (pNETS) showing hypoxia signalling also have a poor prognosis, they paradoxically display a low MVD. We hypothesized that this paradoxon might be explained by alternative splicing of VEGF.
