#4588 Spatial transcriptomics identifies Wnt signalling in multifocal ileal neuroendocrine tumours
Introduction: Ileal neuroendocrine tumours (i-NETs) often present with multiple primary tumours (>30-40%). Recent studies showed no shared somatic mutations in multiple primary tumours, highlighting the need to examine the tumour microenvironment in tumorigenesis. The Wnt signalling pathway is crucial for organ stem cell regulation. While the Wnt/β-catenin pathway influences prostate and pancreatic NET/Cs, its role in i-NETs remains unclear.

Introduction: The Covid-19 pandemic and its restrictions had a negative impact on the general population. Due to lockdowns, food intake increased and exercise decreased. This led to weight gain and changes in body composition. Hospitals reviewed patients (pts) remotely. The effect of the Covid-19 pandemic has not been described in pts with gastroenteropancreatic neuroendocrine tumors (GEP-NETs) treated with somatostatin analogue (SSA).

Introduction: Studies have shown that curcumin regulates malignant tumors through many important signalling pathways, such as Ras, P13K, p53, AKT, mTOR, Wnt-catenin, etc. Regarding the research on the molecular target mechanism of curcumin inhibiting pancreatic neuroendocrine tumors, there is a lack of relevant data at home and abroad. The proven antitumor potential targets of curcumin were intersected with the proteins present in pancreatic neuroendocrine tumor QGP-1 cells, and AKT/mTOR was selected as the central content of this study.

Introduction: Pheochromocytomas and paragangliomas(PPGL) are rare neuroendocrine neoplasms. Up to 25% will develop distant metastases or aggressive behaviour, but there is a lack of accurate risk markers able to stratify patients according to their potential outcome. Genomic characterization of PPGLs has identified 3 clusters: Pseudohypoxic-profile, MAPK-pathway alteration, and Wnt-pathway dysregulation. Alternative splicing dysregulation is emerging as a novel hallmark of cancer, due to its link to oncogenesis and aggressiveness in different neoplasms, including NENs.

#3251 Impact of Wnt signalling on CXCR4 expression and function
Introduction: Loss of Somatostatin Receptor 2 (SSTR2) expression and rising Chemokine Receptor Type 4 (CXCR4) expression are associated with increasing dedifferentiation in Neuroendocrine Tumors (NET). CXCR4 expression is associated with enhanced metastatic and invasive potential and worse prognosis in NET, but might be a theranostic target in NET not sufficiently expressing SSTR2. Likewise, aberrant activation of Wnt/β-catenin signalling may promote a more aggressive phenotype in NET.
